Chokoloskee Island is located at the southern end of State Road 29, approximately 3 miles south of Everglades City.

From Naples / Marco Island

• Take US 41 (Tamiami Trail) East to State Road 29 South
• Follow SR 29 South through Everglades City
• Continue 3 miles south across the causeway to Chokoloskee Island
• Turn left on Mamie Street (or continue to Chokoloskee Drive for the marina)

From Miami / Fort Lauderdale

• Take US 41 (Tamiami Trail) West to State Road 29 South
• Follow SR 29 South through Everglades City
• Continue 3 miles south across the causeway to Chokoloskee Island
